چکیده

background and objective: sorbitol is a non-toxic and slightly hygroscopic compound with different applications. zymomonas mobiles produces sorbitol from sucrose or mixtures of glucose and fructose (formation is coupled with the dehydrogenation of glucose to glucono-δ- lactone). recombinant zymomonas mobilis may produce sorbitol and gluconic acid from glucose and fructose using different divalent metal ions with reduced the ethanol yield and significantly increased yield of sorbitol. current study envisaged to alter the media components, physical process parameters and supplementation of amino acids for enhanced sorbitol production. material and methods: several process variables were evaluated on sorbitol production including carbon sources (glucose, fructose, maltose, sucrose), carbon concentrations (5, 10, 20 and 25 g l-1), nitrogen sources (peptone, tryptone, yeast extract, beef extract and organic nitrogen mix), temperatures (25, 29, 33, 37, 41°c), ph (6, 6.5, 7 , 7.5 ,8), agitation rate (50, 100, 150, 200 rpm) and amino acids (cysteine, cystine, tryptophan)in batch cultivation of lactobacillus plantarum ncim 2912. shake flask cultivation performed under optimum conditions like temperature 37°c, ph 7.0 and agitation rate of 150 rpm, resulted in enhanced sorbitol production. comparative study of sorbitol production in solid state fermentation and submerged fermentation was also evaluated. results and conclusion: batch cultivation under submerged conditions further performed in 7.5-l lab scale bioreactor (working volume 3.0-l) under optimized conditions resulted in maximum cell biomass of 8.95±0.03 g g-1 and a sorbitol content of 9.78±0.04 g l-1 after 42.0 h of fermentation. scale up study on bioreactor resulted in maximum sorbitol yield (yp/x) and productivity of 1.11 g g-1 and 0.50 g l-1 h under submerged fermentation, respectively. conflict of interest: the authors declare no conflict of interest.

Statistical optimization of alpha-amylase production by probiotic Lactobacillus plantarum MTCC 1407 in submerged fermentation.

Production and purification of alpha-amylase by probiotic Lactobacillus plantarum MTCC 1407 has been investigated under submerged fermentation using Mann Rogassa Sharpe medium containing (1%) soluble starch in lieu of glucose (2%) as carbon source. Response Surface Methodology was used to evaluate the effect of main variables, i.e. incubation period, pH and temperature on enzyme production. A f...

Pentose Fermentation by Lactobacillus Plantarum

Extracts of Lactobucillus plantarum derived from cells grown on Larabinose contain an active isomerase, which converts L-arabinose to Lribulose (1, 2), and a ribulokinase, which catalyzes the formation of L-ribulose &phosphate (3,4). Such extracts also catalyze the conversion of L-ribulose 5-phosphate to D-xylulose 5-phosphate, which has been shown to be the key intermediate in pentose fermenta...

Carotenoid production in Lactobacillus plantarum.

Eighteen strains of Lactobacillus plantarum from different origins were screened for carotenoid production, as many of them exhibited a deep yellow pigmentation when cultured as isolated colonies on MRS-agar plates. We found that most of them produced significative amounts of the yellow C(30) carotenoid 4,4'-diaponeurosporene in the range 1.8 to 54 mg/kg of dry cell weight. Although some of the...

High-level production of the low-calorie sugar sorbitol by Lactobacillus plantarum through metabolic engineering.

Sorbitol is a low-calorie sugar alcohol that is largely used as an ingredient in the food industry, based on its sweetness and its high solubility. Here, we investigated the capacity of Lactobacillus plantarum, a lactic acid bacterium found in many fermented food products and in the gastrointestinal tract of mammals, to produce sorbitol from fructose-6-phosphate by reverting the sorbitol catabo...

Production of L-Asparaginase By Fusarium Oxysporum Using Submerged Fermentation

An attempt is made in the present study to optimize the cultural conditions for the production of L-asparaginase by Fusarium oxysporum using submerged fermentation. Different fungal species were isolated and identified using standard manuals, screened initially for the production of extracellular L-asparaginase during their growth on Czapek’s Dox medium containing Lasparagine as sole source of ...

Optimization of bacteriocin production by batch fermentation of Lactobacillus plantarum LPCO10.

Optimization of bacteriocin production by Lactobacillus plantarum LPCO10 was explored by an integral statistical approach. In a prospective series of experiments, glucose and NaCl concentrations in the culture medium, inoculum size, aeration of the culture, and growth temperature were statistically combined using an experimental 2(3)(5-2) fractional factorial two-level design and tested for the...
